In the 'Directory listing filters' dialog box, in the 'Local filters' / 'Remote filters' area, when I click on a label, it doesn't tick the corresponding checkbox. Would it be possible to make these labels clickable in a way like HTML label tags work?

No, not easily. It would interfere with keyboard navigation of that dialog.
